miRNA-27a在人乳腺癌MCF-7/ADR细胞株中的表达及其对细胞耐药的影响Expression of miRNA-27a in human breast cancer MCF-7/ADR cell line and its effect on cell resistance

摘要(Abstract):
目的探讨miRNA-27a对人乳腺癌多柔比星耐药细胞株MCF-7/Adriamycin(MCF-7/ADR)的影响及其可能的作用机制。方法采用real-time PCR(RT-PCR)检测miRNA-27a和P-glycoprotein(P-gp)在乳腺癌细胞株MCF-7及MCF-7/ADR中mRNA表达。将miRNA-27a模拟物(mimics)转染到MCF-7/ADR细胞中,RT-PCR检测转染后miRNA-27a和P-gp的m RNA表达水平。Western blot法检测miRNA-27a模拟物转染后P-gp蛋白表达水平。细胞划痕实验检测转染0、24、48和72 h后细胞的迁移能力。结果 miRNA-27a在MCF-7/ADR细胞中表达量低于MCF-7细胞,差异具有统计学意义(P<0.05)。上调miRNA-27a抑制MCF-7/ADR细胞中P-gp mRNA及蛋白的表达以及细胞的迁移能力,差异均具有统计学意义(均P<0.05)。结论 miRNA-27a可通过抑制P-gp的表达水平逆转MCF-7/ADR细胞对多柔比星的耐药性。
